You can, although the prongs can obscure the image slightly you can still get an idea of whether the diamond has any light leakage or not.
 
No way to use the ideal light(standard lighting environment), so the light you use might not be enough or too much, so need to play around a bit more to get the right environment.

Yes you can get one of those portable led flash lights and backlight the diamond well, with the right angle you should be able to get a decent image. As already stated the prongs and or the setting may get into the way so it is more difficult but not impossible.
